Intruduction:

Expression of the class III POU gene Brn-2, encoding the Brn-2/N-Oct-3 transcription factor, was first identified in mouse brain and is found to be widely expressed in the developing mammalian central nervous system. It has been shown that brn-2 is required for proper retinoic acid induced in vitro differentiation of P19 embryonic carcinoma cells to neural cells and is found predominantly expressed in postmitotic neurons of the mouse neocortex. Recent data demonstrated Brn2 (when combined with Ascl1 and Myt1L) plays a major role in transdifferentiation of human fibroblast cell into neuronal cells. Full-length human Brn2 cDNA was constructed with codon optimization using gene synthesis technology and expressed with a small T7-His-TEV cleavage site Tag (29aa) fusion at its N-terminal and 11 Poly-Arginine (11R) tag at its C-terminal. (This construct deleted 18aa QQQQQQQQQQQQQQQQQQ when compared with wild-type Brn2 protein, NP_005595) It was expressed in E. coli as inclusion bodies. The final product was refolded using our unique “temperature shift inclusion body refolding” technology and chromatographically purified.
